Substrate를 공격하는 nucleophilic residue를 serine으로 하는 Protease이다. 보통 catalytic triad라 하여 serine, histidine, aspartic acid가 main으로 작용하여 scissile bond를 자른다.
Serine protease의 대표적인 Enzyme은
- digestion에 관여하는 chymotrypsin 과 trypsin,
- blood clotting에 관여하는 thrombin이다.
Substrate specificity는 digestion Enzyme의 특성에 맞게 낮은 편인데,chymotrypsin의 경우 scissile bond의 amino region이 bulky hydrophobic residues(Phe or Tyr), trypsin은 basic residues(Lys,Arg), thrombin은 이보다 더 specific하여 Arg-Gly을 인식하여 이 사이를 자른다.
